TetR gene in pBR322 can be cleaved by Bam I and Sal I, while AmpR gene in pBR322 is cleaved by Pst I and Pvu I. If foreign DNA segment is inserted in Pst I site than ampicillin-resistant gene becomes inactive due to insertional inactivation and transformant can be grown on tetracycline containing medium.
